He proposed the 'NAD World' framework linking NAD+ metabolism, the sirtuin SIRT1, and circadian and neuroendocrine control of aging. His lab showed that SIRT1 in the hypothalamus coordinates aging and longevity across tissues, and that the NAD+ precursor NMN and the enzyme eNAMPT can restore youthful signaling in aged mice.
Aging is, in part, a systemic decline in NAD+ — restoring it, and the sirtuin and hypothalamic signaling it powers, is a route to counter age-associated decline across the whole body.
