Multiple RCTs (randomized controlled trials) confirm that oral NMN reliably raises blood NAD+ levels in humans. Evidence for downstream clinical benefits — improved muscle function, insulin sensitivity, and fatigue — exists but is modest, with most trials being small (n<50) and short-term, and few replicating the same endpoint convincingly.
NMN (nicotinamide mononucleotide) is a precursor to NAD+ (nicotinamide adenine dinucleotide), a coenzyme central to cellular energy metabolism, DNA repair, and sirtuin activation. Oral NMN is converted to NMN or NR (nicotinamide riboside) in the gut before entering cells, where it raises tissue NAD+ levels that decline with age.

Most human RCTs are underpowered and industry-funded, limiting confidence in effect sizes. The optimal dose is unclear; 1000 mg/day is on the higher end of studied doses, but dose-response data in humans is thin. Long-term safety data beyond 12 weeks is sparse. NMN's advantage over the cheaper precursor NR (nicotinamide riboside) has not been established in head-to-head trials. Anti-aging or longevity claims rest almost entirely on animal data and have not been demonstrated in humans.
