Bio

Neuroscientist, professor of neurology at Harvard Medical School, and director of the Genetics and Aging Research Unit and co-director of the McCance Center for Brain Health at Massachusetts General Hospital. He co-discovered the first Alzheimer's genes and has spent three decades mapping the genetics of the disease.

Protocol

His team helped find the first genes that cause early-onset familial Alzheimer's — the amyloid precursor protein (APP) and the presenilins — and, through the Alzheimer's Genome Project, later genes like ADAM10 and CD33. His lab built the 3D 'Alzheimer's-in-a-dish' model showing amyloid plaques drive tau tangles, and proposed that amyloid-beta acts as an antimicrobial peptide, tying brain infection and innate immunity to the disease.

Principles

Understand the genes to prevent the disease — Alzheimer's is substantially genetic and begins decades before symptoms, so the path is early detection, a brain-healthy lifestyle, and therapies aimed at the amyloid cascade upstream.
